Abstract

An experimental study was designed to investigate the potential role of amiodarone in chronic theophylline poisoning treatment, compared to propranolol as a standard drug in aspects of arrhythmia, convulsion and gastric erosions. Rats received theophylline (90mg/kg/day) IP for 28 days in theophylline induced toxicity group while the 3 other groups were treated daily with amiodarone (30mg/kg/day), propranolol (1mg/kg/day) or their half doses during the 2 weeks of recovery, respectively to be compared with untreated group. Previous drugs were given to control rats during the last 2 weeks in 2 other groups. On days 1, 14, 28 and 42, serum level of theophylline was analyzed using HPLC method, immediately after dosing in addition to recording ECG and EEG. Finally, serum levels of calcium, potassium were measured, the stomachs were isolated, examined macroscopically to detect the ulcer index then superoxide dismutase (SOD) in gastric tissue was assessed. Also, any abnormal external appearance and/or behavior were checked with mortality rate recording. Treatment by both amiodarone and propranolol showed significant reduction in mean heart rate, improvement of ischemic changes in ECG, EEG morphological changes as abnormal rhythm, spikes and sharp waves especially with amiodarone which showed increase of PSD beta and delta, decrease of PSD alpha while propranolol showed their increase in addition to improvement of metabolic disturbances, increase in mean gastric tissue SOD and the gross examination of stomach of these rats showed minimal scattered petechiae. Amiodarone improved most of the features of chronic theophylline toxicity than propranolol.
